Deficiency of Intact Thrombospondin and Membrane Glycoprotein La in Platelets With Defective Collagen - Induced Aggregation and Spontaneous Loss of Disorder

چکیده
Platelets from a patient with a severe lifelong bleeding tendency. which later spontaneously disappeared, lacked intact thrombospondin and glycoprotein (GP) Ia. Before disappearance of the bleeding disorder, results of coagulation studies and platelet aggregation in response to adenosine diphsophate (ADP). arachidonic acid. thrombin. A231 87, epinephrine. and ristocetin were normal. In contrast. aggregation only occurred in the presence of collagen or wheat germ agglutinin at unusually high doses of these agonists. The platelets adhered normally to purified bovine and human type I collagen. and they did not spread in the presence of methylated type I collagen. No collageninduced clot retraction was observed. Two-dimensional gel

متن کاملRapid purification and partial characterization of human platelet glycoprotein IIIb. Interaction with thrombospondin and its role in platelet aggregation.
Glycoprotein IIIb (also known as glycoprotein IV) is a major glycoprotein present on the surface of human platelets. Recent studies suggest that glycoprotein IIIb may be a receptor site for thrombospondin. Thrombospondin, a multifunctional adhesive glycoprotein released from stimulated platelets, plays an important role in the stabilization of platelet aggregates. In this study, a new method fo...
متن کاملPlatelet adhesion to collagen in individuals lacking glycoprotein IV.
The Naka isoantigen is expressed on glycoprotein (GP) IV (CD36), a platelet membrane GP that has been identified as having a role in platelet interactions with collagen and thrombospondin and in binding Plasmodium falciparum-infected erythrocytes to endothelial cells and melanoma cells. We have studied normal platelets and Naka- platelets from two Japanese donors that have 1% of GPIV by concent...
